Fractionation, Chemical Analysis, and In Vitro Testing Identify Bioactive Components in MC252 Crude Oil

Abstract Crude oil is a complex mixture that includes polycyclic aromatic hydrocarbons (PAHs) as one of its major components. The toxicity some chemically substituted PAHs found in oil, such the methylated species, are relatively understudied. A combination chemical fractionation and analysis coupled with bioassay was used to identify subset activated aryl hydrocarbon receptor (AHR). Silica gel chromatography for primary secondary fractionation, standard reverse phase high performance liquid (HPLC) were final steps. Both gas (GC-) HPLC-coupled mass spectrometry (MS) separate compounds present petroleum fractions. Bioactivity individual fractions identified measured using recombinant yeast strain expressed human (AHRC) transcription factor composed AHR ARNT proteins.
